Walliams has landed the multi-million-dollar lead role in Marmaduke - a family film that is already being hailed as the next Beethoven.
Walliams, 37, will play the dog's trainer Anton. He told the News of the World: "I am absolutely thrilled. The script is hilarious - I shoot it over the summer."
The movie is based on the classic comic strip of the same name, which is about a mischievous Great Dane and the Winslow family.
Film company 20th Century Fox hope there will be sequels. A source said: "They think Marmaduke is going to become the next Beethoven.
"This is a huge opportunity for David to show the world what he can do. Hugh Laurie became a superstar after being in the Stuart Little films. They led to an audition for the series House.
"From doing a family film he's now one of the highest-paid, most popular actors on US TV. David could do the same." Earlier this month we revealed Walliams and Little Britain partner Matt Lucas have put a new series of their BBC comedy on hold while they explore other projects.
Recently David has been spending a lot of time in Hollywood. Just last week he was seen lunching with Jordan there.
A pal said: "He loves the LA lifestyle. He's not quitting the UK altogether but is going to become more focused on the US.
